This dissertation explored the hypothesis that major depressive disorder (MDD) prematurely ages the immune system and central nervous system (CNS). The first empirical chapter, Chapter 2, found that participants with a history of cytomegalovirus (CMV) infection had greater accumulation of late differentiated memory T-cells, but this was independent of diagnosis. Chapter 3 compared the age-related T-cell phenotypes to brain age estimations predicted by a machine learning model trained on gray matter volume data from structural magnetic resonance imaging (MRI) scans. There were no significant correlations between age-related T-cell phenotypes, MDD, or CMV serostatus and brain age estimates. Chapters 4 and 5 explored humoral immunity gained from childhood vaccines in adults with current or remitted MDD. Chapter 4 demonstrated that current or remitted MDD was associated with lower rates of IgG seropositivity for measles. This could mean that MDD-associated immune suppression is disrupting the maintenance of vaccine-derived humoral immunity. Chapter 5 followed up on this finding by measuring antibodies against two other vaccine-derived antibodies against rubella and polio. Polio immunity does not appear to be influenced by MDD, but rubella immunity was lower in the current MDD group. Taken together with the previous chapter's results, MDD does increase the incidence rate of seronegativity to measles or rubella. In sum, our findings provide evidence that MDD is associated with immunological aging, but not brain aging. Furthermore, T-cell aging is influenced by latent CMV infection.
