The place-conditioning assay is thought to measure the rewarding or aversive properties of drug administration. In our experiment, the place aversion assay was used to investigate if the drug combination of MS and an antagonist; either NTX or 6beta-NTXol was aversive. We varied the MS and antagonist doses to capture the range of the CPA assay. The number of conditioning cycles was increased to capture the robustness of the assay. Extinction training was also administered with the intent to measure the resistance to extinction in these groups. These animals were also administered tests of reinstatement and spontaneous recovery. Other dependent variables were measured to monitor various behaviors the animals engaged in during testing. We found that place aversion can be established at the high MS doses of 10MS-0.10NTX and 10MS-0.32NTX. Place aversion was not observed in any of the 6beta-NTXol groups. This aversion can be extinguished after four cycles of extinction conditioning. Further, the highest MS group (10MS-0.32NTX) also demonstrated reinstatement. Spontaneous recovery was not observed in any of the groups. Consistent behavioral changes were not observed in variables that measured locomotor activity and stereotypy. The lack of place aversion in the 6beta-NTXol group lends support to the notion that 6beta-NTXol may produce a minimal withdrawal syndrome than NTX.
