Adipocyte leptin gene expression has been reported to differ relative to stage of the heifer estrous cycle. From estrus to the mid-luteal stage of the estrous cycle, leptin gene expression remains constant. However during the late-luteal stage, leptin gene expression decreases. During this stage Pgf2alpha and its primary metabolite 15-keto-Pgf2alpha are elevated in bovine serum. Therefore, the effect of Pgf2alpha and 15-keto-Pgf2alpha on leptin and PGFr gene expression was quantified in estrogen and progesterone primed adipocytes. Subcutaneous adipose tissue (6 g) and serum was collected from the tail head region of heifers (n=10) at estrus (d 0) and the mid-luteal stage (d 10) of the estrous cycle. Adipocytes were cultured with or without Pgf2alpha or 15-keto-Pgf2alpha (10-11, 10-10, 10-9, 10-8 and 0M; n=3 wells/dose/heifer) and incubated at 37°C for 18h. Leptin and PGFr mRNA from adipose tissue and cells were quantitated using real-time PCR. Serum concentrations of Pgf2alpha and 13,14-dihydro-l5-keto-Pgf2alpha were confirmed by enzymeimmunoassay and estrogen and progesterone were confirmed by radio-immunoassay. Serum concentrations of estrogen and progesterone were greater at estrus and mid-luteal stage of the estrous cycle, respectively. Serum concentrations of Pgf2alpha and 13,14-dihydro-15-keto-Pgf2alpha did not differ relative to stage. Leptin gene expression did not differ in adipose tissue obtained at estrus or the mid-luteal. Fifteen-keto-Pgf2alpha did not affect adipocyte leptin or PGFr gene expression. Physiological concentrations of Pgf2alpha (10-8 M) tended (P=0.1) to decrease leptin gene expression in estrus adipocytes (80.5% +/- 12.7% vs. 0M, respectively). Adipocytes obtained at estrus increased (P< 0.1) PGFr gene expression after PGF2alpha treatment. Mid-luteal adipocytes did not exhibit a similar response to Pgf2alpha treatment. Hence, changes in leptin during the late-luteal stage of the estrous cycle may not be attributed to Pgf2alpha or 15-keto-Pgf2alpha alone.
