The nuclear hormone receptor Steroidogenic Factor -1 (SF-1) establishes and maintains circulating male or female steroid hormone production in vertebrates by regulating cytochrome P450 gene transcription in steroidogenic tissues. We find that the SF-1-like Drosophila nuclear hormone receptor HR39 also plays a role in sexual development. In Hr39 mutant females the sperm-storing spermathecae and its secretory products, as well as the parovaria, are absent or defective, causing female sterility. Spermathecae appear to function as secretory organs, and express genes that likely contribute to sperm maturation as well as storage. Hr39 mutation alters cytochrome P450 gene expression and causes the male-specific courtship gene takeout to be induced in the female reproductive tract. Our results suggest that spermathecae perform functions analogous to the mammalian epididymis, and that steroid signaling pathways act more similarly in controlling mammalian and invertebrate reproduction than previously believed.
