Several studies have shown that HIV-1 infection increases leukocyte integrin expression and collagenase secretion. Based on these observations, it has been hypothesized that HIV-1 infected leukocytes localize in tissues and, subsequently cause tissue-specific pathogenesis. Peripheral blood lymphocytes from transgenic mice showed approximately a 3-fold increase in adhesion to fibronectin, laminin, and type IV collagen, while there was no significant difference in adhesion to type I collagen. Transgenic thymocytes also showed an approximate 3-fold adhesion increase to fibronectin and type IV collagen, and a 1.7 fold adhesion increase to laminin. Transgenic splenocytes showed approximately a 3-fold adhesion increase only to fibronectin. Further, other studies in our laboratory found that thymocytes and splenocytes from transgenic mice secreted an increased level of 92-kDa collagenase. An increase in the level of 92-kDa collagenase was also seen in the serum of transgenic mice. This study shows that, like HIV-1 infected cells, leukocytes from HIV-1 transgenic mice are also more adhesive to extracellular matrix proteins and secrete more extracellular matrix degrading enzymes.
