Mast cells are highly granulated sentinel cells that have been implicated in the pathophysiology of many inflammatory disorders. Through release of granule-associated inflammatory mediators, mast cells play a critical role in initiating immune responses to bacterial infection and in mediating immediate hypersensitivity reactions to allergens. Although much is known regarding the nature of the exocytic responses of mast cells to bacteria and allergens, very little is known regarding the fate of these mast cell-activating agents. We have examined the fate of uropathogenic type 1 fimbriated <italic> E. coli</italic> and particulate allergens in mast cells. Upon binding to their respective receptors, CD48 and IgE/FcϵRI complex, bacteria and allergens were rapidly phagocytosed by mast cells. Both the bacterial and allergen receptors were associated with discrete detergent-resistant membrane domains called lipid rafts where the early signaling events leading to internalization were initiated, however the specific mode of entry and intracellular fate of the cargo internalized via each receptor was distinct. Bacterial entry was associated with the formation of bacteria-sized “caveolae” which completely encapsulated bacteria, forming discrete bacteria-harboring intracellular chambers. Because these chambers failed to fuse with lysosomes, internalized bacteria remained viable within mast cells. Allergen entry involved focal mast cell exocytosis that was immediately followed by the uptake of activating allergen directly into the empty mast cell granule chambers. Because allergens were phagocytosed into granule chambers, they can potentially be re-released upon mast cell stimulation. Our studies reveal that type 1 fimbriated <italic> E. coli</italic> and particulate allergens are phagocytosed by mast cells via two novel mechanisms, which can potentially contribute to the pathophysiology and recurrence of urinary tract infections and asthma.
