Separase is a conserved protease required for the dissolution of cohesion between sister-chromatids at the metaphase to anaphase transition. I found that inactivation of the <italic>C. elegans</italic> homolog by mutation interfered with chromatid separation during embryonic mitotic divisions. Interestingly, affected embryos also appeared swollen <italic>in utero</italic> and displayed a complete failure in cell division, which indicated that separase might have roles in addition to its known role in chromosome segregation. Targeted gene inactivation through RNA mediated interference (RNAi) resulted in a stronger phenotype with a complete failure in chromosome segregation during meiosis I. This is the first study to indicate a role for separase in chromosome segregation in meiosis I in a metazoan.

I compared the pattern of redistribution of a ubiquitous cytoplasmic constituent, mitochondria, in the early embryos of <italic>Caenorhabditis elegans</italic> and <italic>Acrobeloides sp. PS 1146</italic> (<italic>Acrobeloides </italic>), and found them to be dramatically different. These related nematodes share a common early cell lineage, but use different mechanisms to specify the initial antero-posterior axis as well as the order and orientation of cell divisions. In spite of these differences, both species develop into morphologically similar adults. The difference in their mitochondrial redistribution during embryogenesis is an example of a striking evolutionary divergence in a developmental process that ultimately produces similar body-plans.
