A Reversed-Phase (RP) chiral analytical screening system was evaluated to accommodate the varying nature of drug discovery compounds that are submitted for high-throughput separation in the Chiral Laboratory and to stay abreast and up-to-date with current and evolving technology. A refurbished Agilent 1100 Series Analytical HPLC System was released by LAMP (on site instrument maintenance group) for use in this research project. The system is equipped with 2 RP chiral columns from Kromasil, the AmyCoat and CelluCoat. Both performed moderately well in comparison to the normal phase chiral analytical screening system despite system limitations and upgrades. The AmyCoat column separates more chiral compounds sufficiently and to a greater extent than the CelluCoat column based on experimental results. However, this suggestion is highly dependent on the properties of the sample submitted. In addition, the mobile phase and associated additive utilized plays a major role in the enantio-selectivity and retention of the compound.
