Auxin regulates many processes of plant growth and development. Its function depends on its polar transport from cell to cell. Proper functioning of cellular membrane protein targeting is essential for auxin transport. The axr4 mutants of Arabidopsis show a number of phenotypic similarities to mutations in the auxin importer AUX1. Recent evidence shows that AXR4 plays a role in proper targeting of the AUX1 protein (Dharmasiri et al., 2006). We have screened for modifiers of the axr4 phenotype. 45 auxin sensitive axr4-enhancers and 10 axr4-suppressors were isolated in our modifier screen. Here, we report the characterization of one axr4- Enhancer. FE 17.4 is a recessive auxin sensitive axr4-2 enhancer that appears to map to the bottom of chromosome V. It is 50% more resistant in the root to high concentrations of 2,4-D than axr4-2 but is not resistant to 1-NAA which enters cells by diffusion.
