Microbial fuel cells (MFCs) convert organic constituents in wastewater directly to electricity, using a microbial community with exoelectrogenic organisms in an anode chamber connected to a cathode. MFCs offer a promising alternative to traditional biochemical wastewater treatment processes.A suitable inoculum was developed, then initial experiments in a single-chamber MFC confirmed that voltage production was due to microbial activity. A versatile two-chamber MFC was then designed, allowing for the replacement of electrodes and proton exchange membrane (PEM), and facilitating various operations. Preliminary tests in this configuration showed unstable electricity production.Subsequent experiments examined factors including substrate, external resistance (Rex), and MFC configuration. Increasing cellobiose concentration from 0.2 to 1.5 g/L, increased voltage from 81 to 260 mV when Rex was 1000 Ω. The relationship between produced voltage and Rex was determined to be non-linear, however, coulombic efficiency (CE), was found to be a function of Rex, with decreasing Rex increasing CE. Comparisons of MFC performance across different configurations cannot be conducted without a standardized CE.Propionic acid was a more effective substrate than cellobiose or acetic acid. Configuration tests showed that a two-chamber fed-batch MFC with a PEM and a shorter distance between electrodes produced higher voltage. Extended maintenance further improved performance, achieving the highest voltage, 471 mV when Rex was 1000 Ω, power density (PD), 104 mW/m2 , and CE, 23% observed in the study. The microbial community was analyzed using 16S rDNA sequencing, confirming the presence of microorganisms capable of transfering electrons while also showing changes in diversity based on operating characteristics. Continuous mode testing indicated that further research is needed to explore other configurations for continuous operation.
