Polyhydroxyalkanoates (PHAs) are biopolymers synthesized by numerous microorganisms as intracellular carbon storage materials. They are considered a promising and environmentally friendly alternative to petroleum-based plastics. Among the various feedstocks available, lignocellulosic biomass-such as agricultural by-products like corn stover and the organic fraction of municipal solid waste-stands out due to its renewable origin, low cost, and widespread availability, making it an excellent resource for sustainable PHA production. A critical challenge lies in the inherent recalcitrance of lignocellulose: its cellulose-hemicellulose matrix resists most direct microbial assimilation, requiring conversion into monomeric sugars (e.g., glucose, xylose) through sequential pretreatment and enzymatic hydrolysis. This study investigated alkali and acid pretreatment optimization for two biomass types, focusing on reagent selection, process variables (concentration, temperature, residence time, solids ratio) to enhance sugar recovery. Following pretreatment, industrial enzyme formulations facilitated polysaccharide depolymerization. The resulting sugar-rich hydrolysates served as the carbon source for the PHA-accumulating bacterium Pseudomonas putida KT2440, with assessments of substrate compatibility and polymer biosynthesis efficiency. In the subsequent stage of this study, attention is directed toward converting carbon dioxide released during fermentation into a viable input for PHA biosynthesis. Harnessing lignocellulosic biomass as a substrate for PHA generation offers a sustainable and economically feasible route to producing environmentally degradable plastic alternatives.
