Haematococcus pluvialis (Hpw), a freshwater chlorophyte microalga, is the predominant natural source of astaxanthin (ASX), a potent antioxidant known for its anti-inflammatory, anticancer, and muscle pigmentation benefits. Despite its potential, ASX bioavailability from whole Hpw cells is hindered by the presence of a rigid cyst wall. Although disrupting the cell wall can improve bioavailability, the free form of ASX becomes unstable under elevated temperatures, extreme pH, light, or oxygen exposure. To address this, encapsulation strategies have been developed to enhance ASX stability, improving its suitability for functional food products and aquaculture, where natural pigments are preferred for coloring fish flesh. This study investigates the stability of weakened Hpw biomass encapsulated in alginate (ALG) via ionic gelation. The high encapsulation efficiency (EE) (97±2.63%) and loading capacity (LC) (32±0.90%) reported indicate ALG's effectiveness as a wall material for Hpw encapsulation. The resulting ALG-Hpw hydrogel exhibited strong color intensity, which may improve the appearance of feed or functional food products. It also showed favorable physicochemical properties, including a bulk density of 0.59±0.01 g/cm³, moisture content of 11.97±0.20%, and low water activity (0.28±0.00), which helps protect the core material from oxidation. The hydrogels measured 1.30±0.06 mm in size with a sphericity factor of 0.058±0.03, indicating uniformity. CLSM confirmed even Hpw distribution throughout the ALG matrix, while SEM analysis showed surfaces free from cracks, suggesting reduced permeability. Both Hpw extract and ALG-Hpw hydrogels displayed comparable antioxidant activity in the DPPH assay, ranging from 38.32±2.30% to 96.32±0.88% and 33.20±1.55% to 93.30±0.44%, respectively. Encapsulation in ALG improved Hpw's thermal properties by 40.97°C. These findings support ALG as an effective biopolymer for encapsulating Hpw, reducing antioxidant loss and improving thermal properties, which expands its potential in diverse applications. In the second part of the study, ASX-rich extract derived from weakened Hpw was encapsulated in ALG combined with one of five additional polymer wall materials: gum arabic (GA), maltodextrin (MD), pectin (PEC), whey protein concentrate (WPC), or lecithin (LEC). EE ranged from 97.05% to 98.52%, with ALG-WPC-Hpw achieving the highest value. LC remained consistent across all formulations, between 32.46% and 32.95%. Color measurements showed significant variation, with ALG-WPC-Hpw demonstrating the most intense pigmentation, which could boost product appeal. CLSM confirmed uniform distribution of Hpw in all hydrogel formulations. SEM analysis revealed rough textures with varying porosity, all free from visible cracks, while ALG-WPC-Hpw showed some internal air pockets. FTIR results revealed interactions between ASX and the encapsulating materials, confirming successful incorporation. Thermal analysis identified ALG-WPC-Hpw as having the highest thermal resistance. Both free and encapsulated Hpw showed strong and comparable antioxidant activity against DPPH radicals. All hydrogel formulations retained over 96% of ASX at 4°C and over 93% at 25°C during a 28-day storage period, with ALG-WPC-Hpw maintaining the highest retention levels. Among the formulations, ALG-LEC-Hpw demonstrated the greatest ASX bioaccessibility. Overall, the findings highlight the effectiveness of the hydrogels, especially ALG-WPC-Hpw in improving ASX stability, and supporting their potential in food, nutraceutical, and aquafeed applications.
