Protein turnover is an essential cellular process in which old proteins are degraded and replaced with newly synthesized ones. Striated muscle, which includes cardiac and skeletal muscle, is responsible for over 50% of full body protein turnover. Dysregulation of protein turnover is implicated in muscle diseases ranging from sarcopenia to heart failure. And yet, how protein turnover rates differ between different muscle types and disease states remains incompletely understood. We used a combination of stable isotope labeling, proteomics, and imaging techniques to measure the half-lives of hundreds of individual muscle proteins and visualize protein longevity across four different muscle types. We observed muscle-type dependence of protein turnover, visualized protein age heterogeneity at multiple spatial scales, and identified previously unknown long-lived proteins in skeletal muscle. Finally, we applied these approaches to a mouse model of Muscular Dystrophy, where we demonstrated that dystrophic muscles have elevated rates of protein synthesis that can be partially reversed by treatment with a fast skeletal myosin inhibitor.
