Numerous anticancer research outcomes exist but remain distant from effectively combating cancer occurrence and deaths. The situation requires investigation into alternative cancer treatments through medicinal plants. Cancer research needs advancement through ethnopharmacological and ethnobotanical approaches for developing therapeutic agents which produce fewer adverse effects. Several indications show that extracts from herbal plants demonstrate higher effectiveness. Both Azadirachta indica (A. indica) and Vernonia amygdalina (V. amygdalina) have been identified to have anticancer effects by various mechanisms of action. A. indica demonstrates anticancer activity through mechanisms that inhibit cell proliferation while triggering cell growth arrest, differentiation, and programmed cell death. V. amygdalina mechanisms of action involve reduction of cell viability while triggering apoptosis in cancer cell lines. This research distinguishes itself by investigating the impact of A. indica and V. amygdalina combination therapy on HT-29 cell survival and cell cycle regulatory proteins pathways. The plants Azadirachta indica known as Indian Neem and Vernonia amygdalina called Bitter leaf have earned widespread recognition for their multiple medicinal uses. The neem tree originated in Asia and Africa while the bitter leaf exists only in West Africa. The evaluation of their combined effects on HT-29 cancer cells is crucial to determine the safety and efficacy of these traditional therapeutic plants which have extensive applications across the globe. The regulation of cancer-induced changes in cellular regulatory pathways stands as a vital factor for cancer treatment success. Different analytical approaches were utilized in this research to examine Azadirachta indica and Vernonia amygdalina effects on cancer cells through biochemical interactions, cellular toxicity and therapeutic potential evaluation. Overall, our findings indicate that Azadirachta indica and Vernonia amygdalina modulate the expression of Akt, CCND1, and CDK4, suggesting their involvement in the regulation of cell survival and cell cycle progression in HT-29 cells. These results provide an understanding of the molecular mechanisms underlying Azadirachta indica and Vernonia amygdalina's paradoxical effects and their impact on cellular homeostasis, thus suggesting that A. indica and V. amygdalina possess the potential to be anti-cancer therapeutic agents. A thorough investigation of A. indica and V. amygdalina effects on PKB/AKT and its regulatory proteins downstream remains essential.
