The rising global demand for pork has significantly expanded swine production, increasing the use of antibiotic to manage diseases in concentrated animal feeding operations (CAFOs). The use of antibiotics in swine production has been directly associated with elevated levels of antibiotics residues, antibiotic-resistant bacteria (ARBs), and antibiotic-resistant genes (ARGs) in swine manure. A common method for managing swine manure is its application to cropland as a natural fertilizer, which enhances soil quality but also introduces antibiotics, ARBs, and ARGs into the environment. To address these concerns, implementing effective manure management and treatment strategies prior to land application is essential for mitigating the spread of antimicrobial resistance (AMR). This study evaluates the effectiveness of five swine manure treatment strategies- aeration, anaerobic digestion (AD), heat, liming and solid liquid separation (SLS) in reducing AMR. It examines tetracycline (TET)-resistant bacteria, the abundance of TET and macrolide (MAC)-resistant genes, and the concentration of TET and MAC residues across different treatment conditions and timepoints, including both treatment and storage phases, to identify the most effective approach for mitigating AMR in swine manure. The comparative phenotypic results indicate that liming and AD are the most effective treatments for reducing TET-resistant bacteria. Moreover, our genotypic and antibiotic quantification results highlight AD as the most effective treatment for significantly reducing resistant genes (tetO, tetM, and ermB) and total TET and MAC residues in swine manure. This research provides valuable insights for the agricultural sector by offering practical and convenient solutions to reduce AMR spread through manure application, thereby helping to safeguard environmental health and prevent the transmission of AMR to humans.
