Reproductive inefficiency is a primary contributor to the culling of female livestock, and premature culling of livestock results in a tremendous economic burden to producers. Bacterial infections are a recognized contributor to subfertility and infertility in livestock. Mastitis and metritis, evident by gross infection of the mammary glands and uterus, respectively are common reproductive disorders which are known to result in perturbations to fertility. Females that present with overt, clinical illness receive necessary treatment, but the implications of nondetectable infections on female fertility are less well understood. Therefore, the objective of this study was to evaluate the effect of varying low dose LPS endotoxin exposure administered at distinct periods of ovarian follicle development on immune response and reproductive efficiency of affected ewes and of the growth characteristics of subsequent lambs. Ramboulliet ewes (n = 36; 68.2 ± 1.1 kg; age 3 to 7 yr) stratified by body weight and age were assigned to treatment groups. Ewes received subcutaneous injections of saline (CON, n = 12), 1.5 𝜇g/kg BW LPS (LOW, n = 12), or 3.0 𝜇g/kg BW LPS (HIGH, n = 12) on d 5, 10, and 15 of a synchronized follicular wave. Ewes were subsequently placed with a rattle-painted ram on d 16 for a 35-d breeding season. On treatment d 5 and 15, blood samples, peripheral blood leukocytes and rectal temperature were collected before and at regular intervals for 12 h after LPS challenge. Immune response to LPS was confirmed by increased rectal temperature, serum cortisol concentration, and circulating haptoglobin concentration on d 5 and 15. Pro- and anti- inflammatory genes were evaluated by real time polymerase chain reaction. No differences were detected in mRNA expression of tumor necrosis factor or peroxisome proliferator activated receptor gamma. Endotoxin challenge resulted in increased expression of superoxide dismutase 2, toll-like receptor 4, and interleukin-8 on d 15. First service conception rates were 80% for control and 75% for both treated groups. Challenge with LPS resulted in a reduction of lamb birth weight and 60-d adjusted weaning weight. Subacute endotoxin exposure aligning with key ovarian follicle and oocyte maturation events results in a maternal immune response accompanied by reduced production performance in offspring.
