Microfluidic paper-based analytical devices (𝜇𝑃𝐴𝐷𝑠) are a new platform for developing portable diagnostics for resources-limiting settings. These devices enable the detection of various analytes in a sample fluid by accommodating a detection system in a flow guiding pattern fabricated on a paper substrate. Different selection of detection systems requires additional elements on the devices. For example, a colorimetric device may require deposited dye on the device for reporting signal; and an electrochemical device may require screen-printed electrodes for the measurement of signal current. With the cheap and rapid fabrication of such devices, 𝜇𝑃𝐴𝐷𝑠has sparked a rapid development since its introduction, mainly focus on new detection systems and targets, fabrication, and fluid manipulation methods, as well as sample pretreatments and signal amplification. In contrast to the development of additional features in the devices, the device designs are usually done by trail and error-based methods which is both time and resource consuming and can hardly enable optimization the device performances.Such devices can be seen as a chemical product with design frameworks being an active research area in the field of process system engineering in the past two decades. By modifying the framework for addressing the unique design problems in 𝜇PADs, a product design method for designing the 𝜇𝑃𝐴𝐷𝑠 is developed in this thesis. The product design method consists of a conceptual design framework and a numerical platform. In Chapter 2, a numerical platform for predicting the device performances with different designs have been established. In the platform, different imbibition models (Darcy's Law and Richard's equation) are implemented together with solute transport equation with different reaction and source terms, reflecting the selection of different elements in the device. The proposed numerical simulation platform can predict device performances using different combinations of detection system, paper substrate, fluid entrance methods and signal interpretation method. It is also able to simulate devices performances with considerations of partial saturation and provide insights in potential device modifications by sensitivity analysis on various design parameters. In Chapter 3, a conceptual design framework is developed for generating a paper based microfluidics' design, translated from the required detection performance attributes and specifications. The device design will include 4 major elements, which are the required detection tasks to be done in the device to achieve the required detection of the target analyte; the required completion time for the detection tasks specified, the required materials and its amount to achieve the required detection tasks in the specified time; and the physical location, shape, and dimension of the zone where the required detection task will be executed with the specified materials within the specified completion time. By following the framework in a step-by-step manner, a variety of 𝜇𝑃𝐴𝐷𝑠 designs can be synthesized with the required detection specifications, and with their performances predicted by the numerical platform developed in the Chapter 2 for design improvements. In Chapter 4, a systematic analysis of hydraulic properties of different shapes and patterns used in 𝜇𝑃𝐴𝐷𝑠have also been conducted.
