The development of Tissue-Engineered Vascular Grafts (TEVG's) for use in treatment of pediatric cardiovascular disease offers a promising alternative to current synthetic graft models, which typically result in the patient needing additional surgeries to correct for complications related to the inability of these grafts to assimilate into the body. Understanding this potential for TEVG performance improvement, we saw an opportunity in optimizing the scaffolds design. Herein, we first assessed the need for and market viability of TEVGs, creating a needs statement, stakeholder analysis, and market analysis in the form of a Total Addressable Market (TAM), Service Available Market (SAM), and Service Obtainable Market (SOM). We then completed a preliminary chemical composition analysis of six novel TEVG designs, each with varying ratios of Poly-Lactic Acid (PLA):Polycaprolactone (PCL). Upon narrowing our design space to three prototype TEVGs, we conducted an in-depth microstructure analysis to assess the pore size and porosity of the prototype scaffolds. At the same time, we performed an ovine IVC interposition implant study to assess the in vivo development of the TEVG prototypes. This study is ongoing with current results reported. Once complete, this data will be applied to a computational Growth & Remodeling (G&R) model of neovessel development to identify future scaffold designs for optimizing the microstructure, degradation rate, and mechanical profile of the TEVG polymeric scaffold to improve development into a native-like neovessel.
