Fragile X Syndrome (FXS) is a neurodevelopmental disorder that is associated with impaired social behavior and spatial cognition. Both social and spatial cognition rely on the activity of the hippocampus. The hippocampus is believed to support these functions through the activity of place cells, spatially modulated neurons that change their firing patterns in response to environmental changes. The activity of large populations of place cells is modulated by different oscillatory patterns in the hippocampus during specific behavioral states: theta rhythms organize place cell populations during awake exploration, while sharp wave-ripples organize place cell population reactivation during waking rest. In this dissertation, I recorded the activity of hippocampal place cells while rats were exposed to isolated sensory modalities of social stimuli to identify which sensory modality is crucial for the representation of social stimuli in the hippocampus. I found that social odors are the key sensory component necessary for changing place cell activity patterns in hippocampal area CA2, a region known to be important for social coding, in response to social interactions. Further, I recorded the activity of place cells and oscillatory activity in a rat model of FXS (Fmr1 knock-out rats, hereafter "FXS rats") and control rats to determine if the representation of social and spatial stimuli was normal in FXS rats. I found that the representation of social stimuli in CA2 was impaired in FXS rats. I also found that the population-level activity of place cells that represent spatial trajectories was abnormally slow in FXS rats, both during active behavior and rest periods. Together, these results identify specific impairments in the hippocampal representation of social and spatial stimuli in FXS rats and support the hypothesis that hippocampal dysfunction contributes to abnormal social and spatial cognition in FXS.
