As per- and polyfluoroalkyl substances (PFAS), a class of over 12,000 anthropogenic organofluorines, used for industrial/consumer applications, such as water-repellent textiles and food packaging materials, emerge as a contaminant of concern, it is essential to develop methodologies capable of assessing their distribution. Human PFAS-exposure and pollution have been linked to various health concerns, including liver damage and thyroid disease; and their structural resilience and surfactant nature have led to their presence in a myriad of environmental matrices, such as terrestrial and aquatic plants, wildlife, and various waterways. Islands are especially susceptible to PFAS exposure as there is little-to-no barrier between anthropogenic input to fragile aquatic ecosystems. This dissertation conducted the first comprehensive analysis of the PFAS burden on the highly militarized island of Okinawa, Japan, which has a long history of aqueous film-forming foam (AFFF) use and PFAS-contaminated drinking water. The mass spectrometric workflow used to target 31 unique PFAS in Japan was developed and validated by determining the presence and concentration of PFAS in standard reference materials. PFAS quantities determined by the developed method can be used for inter- and intra-laboratory validation checks and improved quality control. Additionally, to streamline data processing of large cohorts of environmental data, a PFAS automated quantitation workflow was developed to rapidly quantify and interpret data. This automated quantitative method was utilized to work up the data of the first statewide PFAS surveillance study in the state of Florida. Employing a comprehensive MS workflow targeting 50 PFAS to more than 2,300 surface water samples throughout Florida, this study aims to establish a baseline of PFAS presence/concentration across the state. In addition, the resultant data will inform communities of high PFAS levels and aid in determining locations for further PFAS investigation.
