The ever-increasing global production of plastics and unrelenting reliance on non-biodegradable petroleum-derived plastics has led to critical issues related to their disposal and persistence in the environment, especially with only a small fraction being considered bioplastics. This thesis addresses these concerns by exploring the production of Polyhydroxyalkanoates (PHA), a family of bio-polyesters that being biodegradable and microbially synthesised from renewable bio-based sources have the potential to replace oil-derived plastics. Amongst the diverse PHAs, Poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) stands out due to its biodegradability and similarities to polyethylene (PE), making it suitable for various applications, including packaging, consumer goods, and biomedical materials. This work is focused on the extremophilic microorganism Haloferax mediterranei, known for its ability to naturally synthesize PHBV in a highly saline environment. To deliver a product with consistent material properties, volatile fatty acids (VFA), specifically C4:0 and C5:0, were utilized as carbon sources. The 3HV fraction of PHBV was previously reported to be proportional to the concentration of C5:0 fed. This thesis is presented in a Journal (Alternative) Format with the novel experimental work performed aimed at improving the production process presented in three separate research articles. In this work, previous fed-batch operations reported in the literature characterized by lengthy fermentations with low productivity, were substantially improved. It was found that maintaining the substrate concentration constant in the media at 0.25 M of carbon delivered optimum results. Implementing a continuously fed fed-batch (CFFB) system quadrupled the productivity from 3.4 mg/(L -1 h -1) in fed-batch to 12.8 mg/(L -1 h -1) in CFFB. A further four-fold increase in productivity was reported when the system was operated continuously; cell density specific PHBV productivity increased from 0.38 mg/(L -1 h -1 OD) in CFFB to 1.43 mg/(L -1 h -1 OD) in the fully continuous operation. A linear greater than six-fold scale up bioreactor from 1.2 to 8 L was achieved resulting in higher polymer production rates. It was proven that by working at a higher temperature, the steady state production rate could be doubled from 82 to 157 mg/h -1. A crucial achievement was the establishment of a robust correlation between substrate consumption and cell density. This correlation simplifies the calculation of feeding rates and concentration in future scale up and higher density operation searching for even higher production rates.Overall, this thesis significantly advances the field of bioplastics by enhancing the controllable synthesis of PHBV via the extremophilic microorganism Haloferax mediterranei. The research presented here not only addresses the urgent need for sustainable plastic alternatives but also paves the way for industrial scale production of biodegradable bioplastics ensuring controllability and improving productivity, ultimately contributing to a more sustainable future.
