Andrographolide is a bioactive compound that is extracted from the leaves and stems of Andrographis paniculata, a medicinal plant commonly used in traditional medicine to treat various ailments. This plant, also known as creat or green chiretta, is an annual herbaceous plant in the family Acanthaceae and is native to India and Sri Lanka. It is widely cultivated in Southern and Southeastern Asia where it has been used for the treatment of bacterial infections and other diseases.The plant grows as an erect herb and can reach a height of 30-110 cm (12-43 in) in moist, shady places. The leaves are lance-shaped and hairless, measuring up to 8 cm (3.1 in) long by 2.5 cm (0.98 in), and the small pink flowers are arranged in lax spreading racemes or panicles. The fruit is a capsule around 2 cm (0.79 in) long and contains many yellow-brown seeds.The production of andrographolide in this plant is affected by various agronomic conditions such as temperature, light, soil fertility, water availability, and plant age. Optimal temperature for growth and andrographolide content is around 30-35°C. The use of supplemental light and appropriate nutrient management practices can enhance andrographolide content in the plant. Appropriate water management practices can also ensure adequate growth and andrographolide production. Harvesting at the appropriate age, generally at the flowering stage, can ensure maximum yield of andrographolide.Andrographolide has been extensively studied for its various pharmacological properties, including anti-inflammatory, anticancer, antimicrobial, antioxidant, and immunomodulatory activities. It has been shown to modulate multiple signaling pathways and regulate gene expression, making it a promising candidate for the development of novel therapeutic agents. Recent research has focused on the pharmacological properties and therapeutic potential of andrographolide.Our study conducted at NCI synthesized various compounds, including isoandrographolide, and tested them for their anticancer activity against 60 cancer cell lines. One of the compounds, 3o, showed the best growth inhibition of 76.90% and 87.54% against leukemia (MOLT-4) and breast cancer (BT-549), respectively. Further testing showed that compound 3g was more active than tamoxifen against MCF-7. Compound 3g arrested cell cycle at G-1 phase of MCF7 and was observed to be inducing apoptosis and loss of mitochondrial potential.
