The Atlantic horseshoe crab (HSC), Limulus polyphemus, one of four extant species of HSC, are an ecologically and commercially important species. In South Carolina, HSCs are harvested for their blood every year as part of a multimillion-dollar industry. Amoebocytes within HSC hemolymph (i.e., blood) can be extracted to create Limulus Amoebocyte Lysate (LAL), a product used to test for endotoxins present in medical devices and injectables. While studies investigating the impacts of commercial bleeding vary widely, mortality estimates of up to 30% have been documented. However, little is known about the factors impacting overall HSC health and mortality. The objectives of this study were to research factors (e.g., age and water temperature) that affect blood chemistry (such as, amoebocyte density and morphology, and hemocyanin concentration and function), metabolism, and mortality of harvested HSCs. HSCs were collected from April to June in 2022 and 2023. In year one, age-related impacts were investigated, and all HSCs were exposed to a temperature of 23 ℃ for one week. In year two, HSCs were acclimated to either 18 ℃, 23 ℃, or 28 ℃ for one week to assess temperature effects. A standard metabolic rate was measured and then HSCs were either mock-bled or had one-third of their blood volume collected, hemolymph that was collected was used for histochemistry and NMR-based metabolomics. Age did not significantly influence blood chemistry, or metabolic rate, but 17 metabolites were significantly different between young and old HSCs. Temperature did not significantly influence blood chemistry. However, temperature significantly influenced metabolic rate and 17 known and 2 unknown metabolites were significantly different across treatments. Results from this study will help us to understand which HSCs are least impacted by harvesting and determine optimal environmental parameters for holding conditions of harvested animals.
