Major depressive disorder (MDD) is a complex psychiatric trait often treated with antidepressant medications. Previous gene expression studies on depression and antidepressant treatment outcome often utilize postmortem brain tissue or peripheral blood. In this novel study design, biopsied living brain tissue from consenting patients receiving clinically relevant deep brain stimulation is sequenced for future research. Using the living brain tissue, this current study aims to compare the differentially expressed gene (DEG) signals between patients with depression and patients taking antidepressants medications. Once characterized by their electronic health records, differential expression analysis and gene enrichment analysis were conducted for both cohorts. A robust DEG signal (π1 = 0.5797, adjusted p-value<0.05) was determined in patients taking antidepressants but not in patients with MDD. Gene ontology analysis indicates enrichment in the immune system, signaling/cell processes and nervous system. These findings extend previous literature findings on DEG and gene enrichment for antidepressant treatment outcome.
