Postpartum depression is one of the most prevalent uniquely female disorders, yet it remains understudied. The sudden decrease in estrogen levels during the postpartum period is thought to contribute to depressive symptoms. Studies using hamsters and rats have shown that estrogen withdrawal following hormone simulated pregnancy (HSP) can induce anxiety-like behaviors. However, the application of this model in C57BL/6 mice to measure anxiety and motivation had not been established. Given that most genetically modified mice are on a C57BL/6 background, this work aimed to further validate HSP to investigate behaviors related to postpartum depression in C57BL/6 mice. Estrogen administration and withdrawal did not produce depression-like effects in C57BL/6 mice, but a significant increase in anxiety-like behavior was observed in the Novel Open Field Test in hormone-treated groups. These results suggest that the model of estrogen withdrawal used here does not reliably induce a depression- or anxiety-like state in C57BL/6 females.
