Highly regenerative tissues such as blood, skin, gut and testis are maintained by the opposing cellular processes of proliferation and cell death. In these tissues, programmed cell death (PCD) functions to remove surplus, damaged or short-lived cells. There are several types of PCD, the most commonly studied of which are apoptosis, autophagy-dependent cell death and programmed necrosis, all of which are regarded as autonomous cell decisions to commit suicide. The resulting apoptotic or necrotic debris are removed by phagocytes that are recruited to engulf the dead/dying cell via recognition of "eat-me" signals, such as phosphatidylserine (PS), exposed on the cell surface. Following engulfment, lysosomal activity is induced to degrade the internalized content. According to this paradigm, only dead cells or cells doomed to die are eliminated by phagocytosis. However, a newly identified type of PCD has been described, named phagoptosis, in which phagocytes engulf and degrade viable cells, thereby leading to their elimination in a cell non-autonomous manner. Although under recognized, phagoptosis is the most prevalent type of PCD in adult tissues.In the Drosophila testis, about a quarter of germ cell progenitors, named spermatogonia, are spontaneously removed by germ cell death (GCD). GCD has been characterized during normal spermatogenesis and under starvation as a non-apoptotic mechanism, which involve lysosomal activity and acidification of the dying germ cells. Hitherto, GCD has been considered as an autonomous PCD pathway.In this PhD thesis, we show that the underlying mechanism of GCD is phagoptosis, in which neighboring somatic cyst cells non-autonomously execute live spermatogonia progenitors. This new role of the cyst cells joins their better-recognized functions as supporters of spermatogonia differentiation. Our results indicate that cyst cells activate the phagocytic machinery and generate endosomal and lysosomal vesicles around live spermatogonia. Targeted germ cells are first acidified, and only then DNA is getting fragmented and PS is exposed. Furthermore, the phagocytic Draper (Drpr) is expressed in cyst cells and the cyst cell membrane penetrates into the targeted germ cells. Importantly, GCD is blocked by knockdown of the endosomal component, rab5, within the cyst cells. Hence, we concluded that the mechanism of GCD is phagoptosis.Additionally, we characterized the roles of distinct Drpr isoforms in GCD during aging. We found that the pro-phagocytic isoform, Drpr-I, is highly expressed in the testis of young males and its levels decline with age indicating on an age-related reduction in phagoptosis. Taken together with the aging-related decline in germ cell proliferation, this data may implicate on a way to sustain spermatogenesis during aging.Lastly, GCD in mammals also deviates from the conventional apoptotic program, and the mammalian Sertoli cells display a dual function of both supporting the developing germ cells and clearing dead cells. Since many functional and molecular features of spermatogenesis are evolutionarily conserved, it is tempting to propose that Sertoli cells may have the capability to perform phagoptosis. The advantage of having a cell non-autonomous rather than an autonomous type of cell death lies in the ability of the tissue to regulate the death rate in response to changing external cues, such as age and nutrient availability.
