Impacts on the head are commonly identiﬁed as the cause of concussive brain injury, which has imposed huge negative health and economic impacts on the world. While brain injury has been recognized as a real issue, researchers are still trying to fully understand how the brain works and how it is injured. Concussive brain injuries occur as a result of a series of fluid-structure interactions (FSI) among the rigid skull, the cerebrospinal fluid (CSF), and the soft brain matter. This dissertation has developed a novel and unique biomimetic approach, with the assistance of modern manufacturing and biological sciences, to examine two fundamental problems in bio-fluid mechanics. (1) the flow and pressurization of the CSF in the subarachnoid space (SAS) and (2) the response as well as the deformation of the brain matter as the head is exposed to sudden external impacts. The goal is to elucidate the critical role of the CSF in transmitting and mitigating external impacts and hence understand the mechanism of concussive brain injury.A systematic, analytical, numerical, and experimental research strategy has been developed. (1)A series of theoretical models for the localized pressure response in CSF have been developed. The models were then verified experimentally. (2)A simplified mathematical model has been developed to capture the motion of the brain matter in response to translational impacts. (3)A novel experimental and theoretical study has been performed to examine the mechanism that governs the motion and deformation of a soft, membrane-bound object, e.g., an egg yolk, bathed in a liquid environment enclosed in a rigid shell. It provides a new perspective for the study of concussive brain injury. (4)A life-like artificial "smart brain" and transparent skull have been developed to examine concussive brain injury. (5) The "smart brain" technique has been applied to the real brain matter (mouse brain and rat brain), which reveals the real brain tissue response to external impacts.As the first study of its kind, this dissertation will significantly improve the understanding of the fundamental mechanisms involved in the incidence and mitigation of brain injury and provide critical insights into head protection mechanisms.
