Tumors exhibit widespread enhancer landscape changes compared to normal tissue which promote disease progression 1-7. These changes are largely attributed to genomic alterations intrinsic to cancer cells8-11. Whether the tumor microenvironment independently contributes to enhancer reprogramming in cancer is poorly understood. Here, by profiling enhancer landscapes on human surgical tissue specimens, we identify a super-enhancer highly enriched in primary colorectal carcinomas (CRC) tumors over patient-matched adjacent normal colon epithelium, that exhibits in vivo context specificity. We demonstrate the super-enhancer and its target gene, PDZK1IP1, are largely silenced in vitro. Xenotransplantation or treatment of CRC cell lines with pro-inflammatory cytokines restores both the PDZK1IP1 super-enhancer and gene expression as well as elicits global enhancer landscape changes that correlate with reprogramming observed in cancer. We establish PDZK1IP1 as a super-enhancer associated oncogene in CRC and show its upregulation alters cancer metabolism to increase oxidative pentose phosphate pathway activity and cellular redox capacity. Collectively, our study demonstrates cell-extrinsic factors within the tumor microenvironment can contribute to enhancer reprogramming in CRC to promote genes upon which cancer cells depend and tumor growth.
