Marine bivalves have an open circulatory system where blood cells (hemocytes) circulate in the hemolymph and wander throughout soft tissue. Most previous studies on bivalve immunity addressed cellular and humoral components of the immune system in hemolymph however, there is a lack of information available on their peripheral immunity, particularly hemocytes present in pallial and extrapallial fluids. Previous work has demonstrated that hemocytes from different body fluids have similar morphological features yet display differences in cell surface epitopes and their functional profiles, including phagocytic activity and reactive oxygen species production. However, little is known about the potential molecular determinants leading to functional differences between circulatory, pallial, and extrapallial hemocytes. To resolve functional specification of the cellular components of different body fluids, hemocytes from the eastern oyster, Crassostrea virginica were first sorted using flow cytometry and submitted to an RNA-Seq analysis of gene expression to describe the molecular characteristics underlying the functional differentiation between granulocytes and agranulocytes from different body fluids. Results generated support the hypothesis that pallial hemocytes might act as sentinel cells like the dendritic cells in vertebrates. More specifically, in this study, functional genes, differentially expressed genes, and gene ontology terms in pallial hemocytes related to capturing and processing antigens, cell migration, cytokines production, and apoptosis inhibition were identified. As for the extrapallial hemocytes, transcriptomic analysis showed unique transcripts and differentially expressed genes involved in shell biomineralization and immune responses. Baseline transcriptomic analysis of pallial and extrapallial hemocytes substantiated the hypothesis that they are a specialized subpopulation of circulating hemocytes. Additional studies are needed to functionally validate the role of identified genes in enabling a sentinel role for pallial hemocytes and shell biomineralization functions in extrapallial hemocytes.
