In this research, the proto-oncogene product, cFos, immunoreactivity served as a marker for stimulated neuronal activity. Using double-label immunocytochemical techniques, the goal of this study was to study LHRH neuronal activation during the estrous cycle in the rat. Then, the relationship between cFos expression in LHRH neurons and the magnitude of LH release was determined. The findings from this research are summarized as follows: (1) During the estrous cycle, LHRH neurons expressed cFos on the afternoon and evening of proestrus but not at other times of the cycle. (2) Pentobarbital, which blocked the preovulatory LH surge, blocked cFos expression in LHRH neurons, but the LHRH neurons expressed cFos on the following afternoon at the time of the expected delayed LH surge. (3) LHRH neurons were activated when the LH surge was induced by exogenous ovarian steroids in ovariectomized adult and intact immature rats. (4) Not all LHRH neurons expressed cFos, even in situations where the cFos stimulation was most dramatic. (5) During the rising phase of the proestrous LH surge, as LH concentrations increased, the percent LHRH neurons expressing cFos increased as well suggesting that the magnitude of the LH surge is regulated by the number of LHRH neurons activated. (6) In the absence of progesterone's action, the decreased surge amplitude was correlated with decreased cFos expression in LHRH neurons (both number of neurons and intensity of staining). In summary, the results from this research demonstrate that LHRH neurons are activated during the proestrous and the steroid-induced LH surges, and further indicate that LHRH neuronal activation plays an active role in the regulation of the LH surge in the rat.
