The rumen microbiome produces VFA and microbial proteins to fulfill nutritional requirements in later life. Therefore, proper inoculation of the fetal gut, which receives contributions from the placental and vaginal microbiomes, may determine lifelong health and metabolic characteristics. In a survey of the placental and fetal microbiomes, amniotic fluid and vaginal swabs were collected from multiparous cows (n = 10) prior to parturition, along with three placental tissues, calf rumen fluid, and meconium upon delivery. Microbial DNA was isolated, purified for 16S sequencing, and analyzed in QIIME2. The meconium and allantois were similar in beta-diversity characteristics, indicating the fetal gut contributes microbes to the allantois through waste elimination. Amniotic fluid was also associated with the fetal gut, which these microbes likely colonize as the fetus swallows amniotic fluid throughout gestation. Like fetal physiology, the fetal microbiome may be subject to the developmental programming of an adverse maternal environment. In an experimental study, a 2x2 factorial design with 70% feed intake restriction and mineral supplementation was implemented with multiparous cows (n = 60). Placental efficiency metrics, placental tissues, and fetal gut contents were obtained, as well as vaginal swabs at 60, 30, and 10 days prior to parturition. The placental microbiome under feed intake restriction was less robust and tended to be less efficient, while mineral supplementation altered the relationship between the fetal gut and vagina. These aberrations indicate the fetal gut is susceptible to programming in utero, and that maternal environment may shape calf health and performance in later life.
