The channel catfish is a well defined model of comparative immunology used to study immune mechanisms of ectothermic vertebrates. This model includes a viable cell culture system, established long-term leukocyte cell lines, and a lineage of homozygous, gynogenetic catfish. A large polymorphic, polygenic family of immunoregulatory receptors exists in catfish termed the leukocyte immune-type receptors (LITRs). LITRS are phylogenetically related to mammalian Natural Killer (NK) cell receptors and some Leukocyte Receptor Complex (LRC) encoded proteins. Two LITR-specific monoclonal antibodies (mAb), CC34 and CC41, were previously generated by immunizing mice with catfish clonal TS32.15 cytotoxic T lymphocytes (CTLs) and the TS10.1 NK cell line, respectively. In this study, LITR-reactive mAbs CC34 and CC41 were used to monitor in vivo cytotoxic responses during viral infection or challenge with Edwardsiella ictaluri. To determine the specificity of LITR-reactive mAbs CC34 and CC41, recombinant (r) LITR proteins were used in immunoprecipitation experiments. Both mAbs recognize conformational epitopes found within the immunoglobulin (Ig) domain 2 and domain 3 (D2-D3) interdomain hinge region of TS32.15 LITR1.1a. Recently, a new LITR, termed LITR622A, was identified and this unique LITR expresses a distinct signal peptide and Ig domain 1 (D1) not previously observed in any other LITR protein sequence. This sequence represents a nonclassical LITR, and it was found to be upregulated in catfish cells by three hours after Channel Catfish Virus (CCV) infection. In addition, to monitor the CC34+ and CC41+ cell response to infection with the intracellular bacterium E. ictaluri, an in vitro model was developed. Briefly, because this pathogen primarily infects pronephros tissue (head kidney, HK) in vivo, HK-derived macrophages were labeled with CellTrace dye and cultured as a platform for infection with E. ictaluri. Autologous peripheral blood leukocytes (PBL) were enriched from whole blood and labeled in parallel using a different CellTrace dye. The PBL were then exposed to the infected macrophages and by day 2 there was a significant increase in CC34+ and CC41+ leukocytes.Combined, these studies demonstrated the utility of the anti-catfish LITR mAbs CC34 and CC41. In addition, the unique LITR622A was identified as a virally-induced stress marker in catfish G14D T cells and Channel Catfish Ovary (CCO) cells.
