Surface-enhanced Raman scattering (SERS), as a versatile technology, has been adapted to a variety of biological studies: identification, detection, tracking and mapping of the target in vivo and in vitro. This dissertation describes approaches towards enabling SERS-based immunoassay for on-site multiplex applications. First, an approach to finely tune an ultrathin shell on the SERS gold nanotags has been optimized, and it improves the sensitivity and specificity of the SERS-based immunoassay. This is of great importance in developing detection of multiple antigens in one single assay. Second, four different Raman spectrally compatible gold nanotags have been synthesized. Given that the SERS-based technology has been demonstrated for its utility in triplex assays for three separate analytes recently, the success of fabricating four different gold nanotags paves the way for a simultaneous quadruplex assay. Third, modification of the recognition biomolecules conjugated on the SERS nanoparticles was proven to be a valid approach to increase the detection sensitivity. Even though the increase was only 1.6 to 3.0 fold, it is worth noting that all the SERS-based immunoassays were performed with a handheld Raman spectrometer. Finally, the SERS-based immunoassay is more than proof of concept, as it has shown its capacity in detecting West Nile virus in avian saliva samples collected in the field with high sensitivity.
