The application of point-spread function engineering to super-resolution fluorescence microscopy is investigated using a spatial light modulator to apply phase modulation to the emission path of a high-resolution microscopy system. Methods for recovering 3D position information from single fluorescent molecules are explored in the context of STORM (Stochastic Optical Reconstruction Microscopy), as is a technique for obtaining spectral information from the fluorescent emitters. The problem of optimizing the signal-to-noise ratio to overcome photon loss in the modulation system is discussed. Representative applications of the developed techniques are considered. General methods for point-spread function engineering using optimization metaheuristics are developed, including a versatile software framework for the design and evaluation of phase modulation patterns.
