Retroviruses are positive-strand RNA viruses that replicate through a DNA intermediate inserted into the host cell genome. Human Immunodeficiency Virus (HIV) is a retrovirus that causes acquired immunodeficiency syndrome (AIDS). AIDS is currently a major worldwide epidemic, and although some treatments are available that can help treat the infection, there is neither a cure nor a vaccine.

The lack of a cure or a vaccine is due in part to the fact that the cell biology of retroviral infections has remained elusive. Most models of retroviral infections are based on principles of retrovirology and do not consider the biology of the host cell.

One organelle in host cells, called the endosome, can form inwardly budding vesicles containing cytoplasm from the cell. If an endosome contains multiple vesicles, it is called a multivesicular body (MVB), and can fuse with the plasma membrane. This fusion event results in the release of the vesicles into the extracellular milieu, and these vesicles are called exosomes. Exosome biogenesis can also occur directly at the cell surface when endosomes or MVBs fuse with the plasma membrane. Once released, other cells can take up exosomes, resulting in a cellular pathway of intercellular vesicle trafficking.

The first part of this work describes the Trojan Exosome hypothesis, which states that retroviruses utilize the exosome biogenesis pathway for the formation of retroviral particles and the exosome uptake pathway as an alternative mode of infection. The second part of this work is an extension of the Trojan Exosome hypothesis and describes an alternative explanation for the evolution of alloimmunity and the genesis of adaptive immunity. Finally, I describe my laboratory work which provides experimental support for the Trojan Exosome hypothesis.
