Critical limb ischemia and Duchene muscular dystrophy are two devastating diseases that eventually result in necrosis and ischemia of skeletal muscle. Minorities, particularly African American women, are at a higher risk for critical limb ischemia for reasons unrelated to social status. The amputation and mortality rate of this disease are substantial. Duchene muscular dystrophy, however, is an X-linked recessive genetic disorder that affects young boys that eventually leads to their death in their early twenties. Ongoing clinical trials are using bone marrow cells and endometrial regenerative cells as a means of stimulating angiogenesis; however, efficiency of using these cells to repair necrotic myofibers is unknown. This study shows that pharmacological modulation of muscle-derived stem cells implanted into the gastrocnemius of an ischemic or dystrophic mouse model may in some cases stimulate angiogenesis and early muscle repair, as well as reduce fibrosis, but this is presumably counteracted by myostatin over-expression.
