The segregation of the three primary germ layers during gastrulation is one of the first critical cell fate decisions in establishing the embryonic body plan. The definitive endoderm gives rise to the epithelial lining of the digestive and respiratory systems and its associated organs. Although studies in model organisms have revealed that a conserved molecular program orchestrates endoderm formation, our understanding of endoderm formation still remains elusive, especially in humans.

Human embryonic stem cells self-renew and have the potential to differentiate into all the cell types of the three embryonic germ layers, making them an attractive source of cells for regenerative medicine. Understanding definitive endoderm formation in hESCs will not only serve as a valuable model for human development but also help progress the in vitro differentiation of embryonic stem cells for potentially useful cell based therapies and drug discoveries. In this dissertation, we describe a novel and efficient method of generating definitive endoderm using FGF2 and LiCl and the molecular mechanisms involved.
