Previous research in our lab has shown that there is a transmission ratio distortion (TRD) due to reduced fertilizing ability of the translocation-bearing sperm in males heterozygous for Rb(6.16) or Rb(6.15)translocation. The murine Sperm Adhesion Molecule 1 (Spam1) gene located at the proximal region of mouse chromosome 6 is an attractive candidate for the sperm dysfunction, since it plays an important role in fertilization. A decreased mRNA and protein level of this testis-specific gene was found in Rb-bearing mice, further supporting its candidacy.

520 $a Expression analysis shows that Spam1 is a haploid-expressed gene and its transcripts and protein were shown to be compartmentalized in the cytoplasm of spermatids. This finding indicates that the Spam1 product is not shared among spermatids via the cytoplasmic bridges as generally expected of spermatogenic genes. Consequently, phenotypically unequivalent sperm are produced in heterozygous Rb-bearing males and this leads to TRD.
520 $a The murine Spam1 was shown to be encoded by four exons covering 10.5 kb, similar to the human homologue. The promoter region of Spam1 was shown to contain an 8-nucleotide CRE sequence (TGATGTCA) at $- $5 7. Gel shift assays reveal that the putative CRE can bind to purified CREM (cAMP response element modulator) or endogenous CREM in testis nuclear extract, and that the binding is sequence-specific. In vitro transcription analysis demonstrates that the CRE sequence is required for the transcriptional activity. Additionally, Spam1 transcripts were shown to be absent in CREM knockout mice.
520 $a Expression analysis of other genes located near the translocation junction shows that the lower level of Spam1 is not due to a silencing effect of the prolonged centromeric heterochromatin at the translocation junction. No gross alteration of the gene structure has been found in the Rb mice. However, as many as 11 point mutations were found in the promoter, 5 $\ sp\prime $ and 3 $\ sp\prime $ UTR, as well as the coding region in Rb(6.16) homozygotes. It is proposed that these mutations are in linkage disequilibrum with the translocation junction due to the suppression of recombination. One or more may affect the transcription rate or the stability of mRNA and account for the lower steady-state level of Spam1 in Rb-bearing mice.
