Techniques utilizing Matrix-assisted laser desorption ionization mass spectrometry (MALDI-MS) for the analysis of molecules of biological and environmental importance are described in this dissertation. The implementation of new MALDI-MS techniques resulted in rapid sample analysis, reduction in sample preparation steps and low detection limits.Subjects--Topical Terms:

The MALDI mass spectra of several of the most commonly prescribed benzodiazepines including alprazolam (Xanax) and diazepam (Valium) have been acquired in both positive and negative mode with a variety of MALDI matrices. Through the use of an internal standard, calibration curves have been constructed which demonstrate the quantitative nature of MALDI analysis for these compounds even when multiple benzodiazepines are present in a single sample.

Qualitative analysis of pesticides by direct coupling of Thin Layer Chromatography (TLC) with MALDI was demonstrated. TLC/MALDI takes advantage of the high sensitivity of MALDI while utilizing the relatively quick and inexpensive separation offered by TLC. The optimization of a protocol to analyze cationic pesticides by TLC-MALDI is reported and tested on normal phase, reverse phase, and cellulose TLC plates.

9-aminoacridine (9AA), a small molecule with basic character, is introduced as a negative mode matrix for several types of compounds. Pesticides, drugs of abuse, and bile acids, which were difficult to ionize using typical MALDI matrices, were the primary focus of this investigation.

Finally, a novel method for the determination of the herbicides diquat and paraquat in water samples utilizing MALDI has been developed. Solid phase extraction cartridges, which consist of a glass micofibre disc impregnated with reverse phase material (C-18) encased in a polyethylene housing, are specially prepared for use in the ion-pair mode. The sample is applied under full vacuum, followed by addition of a MALDI matrix solution. The disk is dried, removed from the cartridge holder and analyzed directly by MALDI-MS. The use of an internal standard, applied to the disc, permits this method to be quantitative as well as qualitative.
