The ascomycete fungus Trichoderma virens (strain G20) has been used as biocontrol agent to suppress soilborne microorganisms that are pathogenic to plants. It has been reported that antibiotic peptides produced by T. virens which play important roles in its biocontrol activities. Among them, gliotoxin involves in the suppression of Pythium-induced damping-off; siderophores are involved in iron uptake; peptaibols exhibit anti-bacterial and anti-fungal properties. Small peptides are often made nonribosomally by a class of enzyme called peptide synthetases (PPS).Subjects--Topical Terms:

This dissertation is directed at characterizing PPS genes of T. virens. Degenerate oligonucleotides based on PPS adenylation (A) motif were applied for PCR screening. Nine unique PPS A-domain p4, p12, p16, p22, p32, p38, p42, ser2, and ser15 were identified. Long range PCR was applied to link these individual domains into different PPS. P8, p32 and p2 are in the same gene. Total 12.5 kb of this gene have been sequenced and multiple disrupts indicate that this PPS is associated with the biosynthesis of an 18-mer peptaibol, but not other peptaibols. By cosmid library screening cos16, harboring PPS16, which includes p16, was identified. About 14kb of PPS16 has been sequenced and the peptide product of this 14kb DNA fragment has been predicted to produce a partial peptide of D-ala-Val-Pro-Leu/bmt. A-domain clone ser2 is related to the siderophores PPS in fungus Neurospora crassa.

Attempts to clone PPS genes also resulted in the isolation of cosmid cos32. Cos32 was able to specifically complement the gliotoxin non-producing UV mutant, m37, of T. virens. The 37kb insert of cos32 includes a putative multiple drug resistance, catalase/peroxidase, ABC transporter, and other 17 ORFs. These identified genes may provide a molecular function to the defect that prevents gliotoxin biosynthesis in m37 and a link to other genes involved in its biosynthesis.

In conclusion, I was able to identify multiple A domains and at least three different multi-module PPS genes in T. virens. This is important since multiple PPS genes have not be previously characterized in a single fungal genome. A role for each PPS gene, or at least associated genes, is evident for each of the peptide metabolites known to be made by this fungus. One novel PPS, PPS16, appears to be related to an uncharacterized peptide whose partial sequence can be predicted.
