Cell-cell recognition is a key event that determines the activation of T lymphocytes, which are the orchestrator of the adaptive immune system. The immunological synapse, a macroscopic spatial pattern of receptors and ligands which forms in the intercellular junction between T lymphocytes and antigen presenting cells (APCs), is implicated in information transfer between these immune cells. Based on a statistical mechanical model developed by Qi et al. (Qi, Groves et al. 2001), we will discuss the mechanisms that underlie the formation of different synapse patterns at different stages of the life cycle of T cells. The functional consequences of synapse formation to T cell activation are explored. We also develop a numerical simulation method to study intracellular signaling cascades which is coupled to synapse formation. Appling this algorithm, we seek possible explanations for some controversies regarding T cell signaling and activation.
