Tooth eruption is a multifactorial process that involves the movement of existing tissues and the formation of new tissues coordinated by a complex set of genetic events. We have used the model of the un-opposed rodent molar to study morphologic and genetic mechanisms of tooth eruption. Following the extraction of left maxillary molar teeth in mice, mandibular molars were allowed to super-erupt. The mice were injected with fluorescent dyes. Mandible tissue blocks were prepared for ultrathin sections and fluorescent microscopy. Other specimens were prepared for RNA extraction, microarray analysis, and immunohistochemistry. Labeled tissue sections revealed significant differences in bone and cementum apposition between the experimental and the control side. Microarray analysis demonstrated significant increase in gene expression of collagen I, integrin beta5, and SPARC. The results from the microarray were verified using RT-PCR. The immunohistochemistry revealed increased levels of expression in the periodontal ligament of the un-opposed molar.
