Activated sludge systems are used to treat wastewater in many parts of the world. With total suspended solids (TSS) concentrations up to 10 g/L and a versatile mixed culture of microorganisms, they present an ideal system for reduction of estrogens by sorption and microbial metabolism.

A yeast construct transfected with human estrogen receptor (ERalpha) was used to adapt a sensitive yeast assay and develop a protocol to measure estrogenic activity in wastewater. The assay combined detection of estrognic activity to as low as 0.03 ngE2/L and greatest quantitative sensitivity between 250 and 2900 ngE2/L.

Batch experiments were carried out with sludge solids concentrations ranging from 1 to 5 gTSS/L and E2 spiked doses of 100 ng/L, 100 mug/L and 20 mg/L. The assay was used to measure reduction of total estrogenic activity from the sludge supernatant over a 15-day period after which the sludge was re-spiked with E2 and further monitored to check for reduction by pre-exposed sludge. The reduction was generally increased by increasing sludge solids. For 100 ngE2/L there was >40% reduction of estrogenic activity within 15 days which improved to >70% by pre-exposing the sludge. The estrogenic activity produced by a dose of 100 mug/L was readily removed within 15 days. However, re-spiking the sludge with the same concentration caused some sludges to lose reduction capacity. The same observation was made at 20 mg/L dose where although up to 70% reductions could be obtained with >2 gTSS/L, only some sludges showed improved reduction after re-spiking.

Keeping the sludge aerated and viable by mixing was found to enhance reduction. This was confirmed using static control experiments and sterile sludge, and by comparing short-term solid versus liquid phase partitioning. Unmixed sludge removed estrogenic activity slower and only solids concentrations <1 gTSS/L maintained estrogenic activity reduction capacity, probably due to low oxygen demand.
