We then consider the use of pedigree data methods only to explore association between ancestry and longevity in individuals of Ashkenazi Jewish (AJ) ancestry. We evaluated the relation between AJ ancestry and all-cause mortality rates among 37,362 Caucasian men and women from Australia, Canada and the United States. We estimated death-rate ratios (DRR's) in subjects with one, two, three or four AJ grandparents relative to those among individuals with no AJ grandparents. After adjusting for year of birth, country of residence, age at risk of death, and educational level, we found that death rates among individuals with four AJ grandparents were significantly lower than those among individuals with no AJ grandparents. Death rates were lowest among individuals of mixed heritage. The nonlinear relation between death rates and number of AJ grandparents suggests that the differences in longevity may reflect socioeconomic or lifestyle characteristics unrelated to educational level, rather than genetic factors.

When allele frequencies at any two loci are different in the ancestral populations, there is increased gametic association between these loci in the admixed population. Admixture mapping methods that require the estimation of ancestry at specific genetic markers can be used to exploit the resulting disequilibrium to identify genes underlying traits that differ qualitatively or quantitatively between populations. We propose an affected-only test for linkage in admixed populations based on the principal of admixture mapping and explore its properties in the case where individuals have both DNA and pedigree data available. We compare our test to the Transmission Disequilibrium Test in admixed populations. Results suggest that, in populations where parental ancestral heterozygosity at the marker loci under study is greater than 0.5, our test will outperform the TDT.
