To test the hypothesis that ovarian hormones alter exercise metabolism away from blood glucose and toward free fatty acids (FFA), we examined the effects of endogenous (menstrual cycle fluctuations) and exogenous (oral contraceptives (OC)) ovarian hormones on peak oxygen consumption. (V˙O2peak) and fat mobilization ([1,1,2,3,3-2H]glycerol rate of appearance (Ra)) during 90 minutes of rest and one hour of mild (45% V˙O 2peak) and moderate (65% V˙O2peak) intensity exercise. Eight moderately active female subjects (24.8 +/- 1.2 yr), with consistently normal menstrual cycles (24--32 days) were examined during the follicular (FP-days 3--9) and luteal (LP-days 18--24) phases of the menstrual cycle before OC and during the inactive (IP-no hormone ingestion) and high dose (HP-0.035 mg ethinyl estradiol and 0.25 mg norgestimate) phases after 4 months of tri-phasic OC.

During peak exercise before OC, there were no significant differences between FP and LP in any of the variables studied. There were also no differences between IP and HP with OC. Dietary composition, exercise patterns and peak heart rate, pulmonary ventilation and respiratory exchange ratio did not change with OC use. However, OC use significantly (p ≤ 0.05) increased body weight (59.6 +/- 2.3 to 61.2 +/- 2.6 kg) and fat mass (13.3 +/- 1.3 to 14.5 +/- 1.3 kg), and decreased V˙O 2peak (-11%, 2.53 +/- 0.21 to 2.25 +/- 0.18 l·min -1).

There were no significant differences in glycerol Ra between FP and LP at rest or during exercise. While there were no differences in resting or exercise FFA (except a lower concentration during 65% V˙O2peak at IP) and glycerol concentrations or whole body respiratory exchange ratio between phases, OC use significantly (p ≤ 0.05) increased glycerol R a during exercise at both 45% and 65% V˙O2peak. This increased fat mobilization with OC use was associated with higher cortisol concentrations at rest and during both intensities of exercise.

In conclusion, these results suggest that in moderately active young women, (1) endogenous ovarian steroids have little effect on V˙O 2peak or fat mobilization, but (2) the exogenous ovarian steroids in OC decrease V˙O2peak and (3) increase fat mobilization during exercise.
