How an organism, or in a simplified case, a cell, senses changes in the environment, and generates responses that improve its chance of survival, is a fundamental question in biology. In budding yeast Saccharomyces cerevisiae and its pathogenic relative, Candida albicans , the Rim101p pathway plays a key role in cellular responses to changes in the environment. Previous and current studies have shown that Rim101p activity is required for adaptation to environmental pH and ion concentrations, morphological switches, the decision to enter meiosis and to sporulate, and host-pathogen interactions.

520 $a Rim101p is a zinc finger transcription factor that is activated by a proteolytic cleavage that removes &sim;100 C-terminal residues. The processing of Rim101p is stimulated by alkaline pH, and requires gene products of RIM8, RIM9 and RIM13. In this study, I focused on the molecular mechanism of Rim101p processing. I identified anew Rim101p pathway member, Rim20p, which interacts with Rim101p and is required for its processing. I further demonstrated that Rim20p interacts with Snf7p to form a scaffold that recruits the putative protease Rim13p to cleave Rim101p. More intriguing is my discovery that eight ESCRT proteins of the MVB pathway, including Snf7p, are required for Rim101p processing, suggesting an intrinsic relationship between the two pathways.
