Scutellaria baicalensis Georgi is a medicinal plant used for treatment of various ailments and diseases. It is ubiquitous throughout eastern Asian medicine with potential for commercial production in the Western countries. The secondary metabolic makeup of S. baicalensis consists mainly of flavonoids, especially in roots, with the human medicinal effects being attributed to the vast bioactivities of the flavonoids. A method of flavonoid analysis was developed and partially validated for S. baicalensis in root and shoot tissue including quantification of eight flavones and tentative identification of two additional flavones using HPLC-DAD-MS. Subsequent analyses of S. baicalensis, including baicalin and related flavones, resulted in total flavonoid mass contents of 0.65% and 0.93% in cell line 111 in vitro shoot and root tissue, and 0.54% and 11% in greenhouse shoot and root tissue, respectively. This method proved accurate and precise for the analysis of various S. baicalensis plant types.
