The purpose of this research was to develop and optimize immediate release tablets of the drugs Glipizide (5 mg) and Metformin hydrochloride (500 mg) alone and in combination. The effect of the excipients Lactose, Cab-O-Sil, Kollidone K90, and Magnesium Stearate on the tablet release rate were also characterized. More specifically, the state and interactions between the drug and excipients were characterized by X-ray diffractometry (XRD), Differential thermal analysis, differential Scanning Calorimetry (DSC) and thermogravimetric analysis (TGA). XRD analysis showed that drug alone (Metformin), (Glipizide) and final product were crystalline in nature. UV analysis showed that there was no physicochemical modification in the solid dispersion formulations as compared to final product. Thermogravimetric analysis suggested that Metformin and Glipizide can partially form a solid solution as the temperature increases. The appearance of an exothermic peak on DSC thermogram suggested corresponding decomposition of the drug mixture and solid dispersion. Dissolution studies (n=6) of the tablets of drug alone were conducted in 900 mL simulated intestinal fluid of 0.2N HCL with USP apparatus. (Abstract shortened by UMI.)
