Pseudomonas aeruginosa (P. aeruginosa) keratitis is a sight threatening disease that is mostly associated with extended contact lens wearers or immunocompromised patients. Our group has earlier published the importance of human tear fluid in protecting corneal epithelial cells against P. aeruginosa infection. The work here, therefore, aimed at identifying the responsible tear component(s) and became the first to report that surfactant protein-D (SP-D) is present in both the human tear fluid and the cornea. We further characterized SP-D expression and explored the molecular mechanism of the regulation of SP-D production in human corneal epithelial cells in response to P. aeruginosa exposure.Subjects--Topical Terms:

520 $a First, we sought to identify specific tear component(s) that can confer protective effect against P. aeruginosa. Our results showed that SP-D is present in human tear fluid and involved in its protection against P. aeruginosa invasion. Western blot, RT-PCR and immunofluorescence microscopy showed that cultured human and mouse corneal epithelial cells are able to express and secrete SP-D.
520 $a Next, we demonstrated that the expression and secretion of SP-D by corneal epithelial cells are subject to regulation upon exposure to whole bacteria or bacterial components. Cultured human corneal epithelial cells challenged with heat-killed P. aeruginosa, or P. aeruginosa lipopolysaccharide or flagellin protein (FliC) showed increased expression and secretion of SP-D. This upregulation is MyD88- and NF-kappaB-independent, but JNK-, ERK- and p38-dependent. Bacteria expressing mutated FliC composed flagella, which are defective in TLR5 binding site, induced an even stronger SP-D response than bacteria with normal flagella did. Taken together, P. aeruginosa-induced SP-D and pro-inflammatory cytokine IL-8 upregulation appeared to be mediated by separate signaling pathways.
520 $a In summary, the work presented here has demonstrated that SP-D plays a significant role in ocular defense against P. aeruginosa and has shed some light on the regulation of SP-D expression at the molecular level. The findings may allow development of novel preventive methods and better therapeutic treatments for P. aeruginosa keratitis.
