This dissertation examines the biological actions of cyclopentenone (A 2/J2) isoprostanes, reactive lipid peroxidation products formed in vivo under conditions of oxidative stress. Mass spectrometric evidence is presented demonstrating that cyclopentenone isoprostanes are formed abundantly in rodent and human brain tissue, and are increased under condition of oxidant injury. Experiments employing synthetic cyclopentenone isoprostanes (15-A2- and 15-J2-IsoPs) reveal that these compounds are highly neurotoxic, and induce neuronal apoptosis at biologically relevant concentrations via activation of discreet, redox-sensitive signaling pathways. Cyclopentenone IsoPs also potentiate neuronal death in several models of neurodegeneration. Further data is included showing that these compounds can also modulate the inflammatory response via inhibition of the NF-kappaB pathway in macrophages. This dissertation provides evidence that cyclopentenone isoprostanes are highly bioactive lipid peroxidation products, and may mediate some of the pathogenic effects of oxidative stress.
